Abstract

Objective: To study the effects of matrine on proliferation and apoptosis of K562 cells.

Methods: Cell culture and cell count, cell cycle phases and number of apoptotic cells were analyzed by flow cytometry. Morphology of apoptotic cells was studied by electron microscopy, BCL-6 and proliferating Cell nuclear antigen (PCNA) were checked by Western blot.

Results: Matrine could inhibit the proliferation of K562 cells with inhibition rate of 69% after 72 hours matrine induction; number of K562 cells increased significantly in G1 phase, and number of apoptotic cell was 68% at 5th day of matrine treated cells; the typical morphology of apoptotic cells of K562 cells was observed by electron microscopy; BCL-6 was increased and PCNA was decreased in K562 cells induced by matrine.

Conclusion: Matrine inhibited the proliferation and induced the apoptosis of K562 cells significantly, the mechanism of which might be related to cell cycle arrest and the change of PCNA and BCL-6.
