A loss of glycerol export from adipocytes causes alterations to the triglyceride/fatty acid cycle and perturbations in glucose homeostasis. The phenotype of young Aqp7-/- mice is relatively mild, characterized by fasting-induced hypoglycemia. In older mice, a loss of glycerol transport results in a dramatic accumulation of triacylglycerol in adipose tissue and metabolic alterations, including elevated FFA, hyperglycemia, and insulin resistance. Red arrows indicate metabolic perturbations in Aqp7-/- mice or in mice with reduced expression of Aqp7 in 3T3-L1 adipocytes. [Modified from Reshef et al. (6).]
