Proposed scheme for regulation of Aβ degradation by PKCε via ECE. PKCε activates the transcription factor Ets-1, which promotes ECE expression. ECE cleaves isoforms of big ET (themselves the products of pre-pro-ET processing by furin) to produce active ET isoforms (ET-1, -2, and -3). ECE also degrades Aβ (including both Aβ1–40 and Aβ1–42), produced through the action of α-, β-, and γ-secretases, into smaller fragments; ECE cleavage sites occur between Aβ residues 16 and 17, 17 and 18, and 19 and 20 (7).
