Erythrocyte and plasma fatty acid patterns in dogs with atopic dermatitis and healthy dogs in the same household
Abstract
Recent studies have indicated that dogs with canine atopic dermatitis (CAD) may have a disorder of fatty acid metabolism: possibly low or absent activity of delta6-desaturase or delta5-desaturase, or both. To clarify this possibility, we examined the erythrocyte and plasma fatty acid patterns of 8 dogs with CAD and their 8 healthy housemates. Atopic dermatitis was diagnosed according to the criteria proposed by Willemse; other causes of dermatitis were excluded clinically and by appropriate tests. Erythrocyte ghosts were prepared from blood samples. Membrane lipids were extracted and separated by thin-layer chromatography. From plasma and lipid fractions, fatty acid content was determined by gas chromatography. In erythrocytes, but not in plasma, we observed significant differences in the fatty acid pattern that suggested a reduction in the n6 fatty acid products of the delta6- and delta5-desaturases in dogs with atopic dermatitis when compared with healthy housemates.
Des études récentes rapportent que des chiens atteints de dermatite atopique canine (CAD) pourraient avoir un désordre du métabolisme des acides gras, une activité réduite ou absente de la Δ6-désaturase ou de la Δ5-désaturase, ou les deux. Afin de clarifier cette situation, nous avons examiné les érythrocytes et les patrons d’acides gras de huit chiens souffrant de CAD et de leurs huit compagnons normaux partageant la même maison. Une dermatite atopique a été diagnostiquée en fonction des critères proposés par Willemse; les autres causes de dermatite ont été exclues sur la base des signes cliniques et des tests appropriés. Des érythrocytes fantômes ont été préparés à partir d’échantillons sanguins. Les lipides membranaires ont été extraits et séparés par chromatographie en couche mince. À partir du plasma et des fractions lipidiques, les concentrations d’acides gras ont été déterminées par chromatographie en phase gazeuse. À partir des érythrocytes, mais pas du plasma, nous avons observé des différences significatives dans les patrons des acides gras qui étaient suggestives d’une réduction des produits n6 des acides gras issus des Δ6- et Δ5-désaturases chez les chiens souffrant de dermatite atopique comparativement à des chiens cliniquement normaux logés sous le même toit.
