TH signaling in RTH. (A) In normal individuals, TRH triggers TSH release, which stimulates TH production. Circulating TH binds TR (green), represses the TRH and TSH genes, and exerts positive (green) and negative (red) effects on genes in peripheral tissues. (B) In GRTH, mutated TR (red) is unable to transduce TH signals. Negatively-regulated genes are derepressed (green) and positively-regulated genes are suppressed (red). (C) In CRTH, mutated TR (yellow) is defective in negative regulation. TRH and TSH are derepressed, and TH production is elevated. Although negatively-regulated peripheral genes are derepressed positively-regulated genes remain correctly regulated.
