Association of Poldip2 with Nox4 increases ROS generation and strengthens focal adhesions and stress fiber formation in SMCs. The dissociation of Poldip2 and Nox4 at the trailing edge of a migrating cell is predicted to release adhesions, whereas the association of Poldip2 with Nox4 at the leading edge increases adhesions. The dynamic regulation of focal adhesions by Nox4-Poldip2 may be important for directional cell migration. The role of Nox4-Poldip2 in the nucleus is unknown.
