A simplified model of insulin-stimulated cardiac aging. Caloric intake results in pancreatic insulin secretion and insulin binds to cardiomyocyte cell surface receptors. Insulin binding to cognate receptors results in activation of an intracellular signaling cascade that includes IRS-1, PI3K, PDK1 and Akt family members. Akt proteins phosphorylate and inhibit FoxO transcription factors by promoting their nuclear exclusion. Akt proteins also phosphorylate and inhibit TSC2, ultimately leading to activation of TOR complex 1 via Rheb. FoxO transcription factors promote autophagy whereas TORC1 inhibits autophagy. Autophagy prevents lipofuscin accumulation and aging of the myocardium.
