Simplified diagram showing how the bone–pancreas axis may influence energy metabolism. Insulin interacts with the osteoblast through an insulin receptor (IR) to produce osteocalcin. Osteocalcin is activated to undercarboxylated osteocalcin, perhaps by a low pH microenvironment of the bone undergoing resorption, which then interacts with β cells of the pancreas to release insulin to influence energy metabolism. The molecular mechanism of undercarboxylated osteocalcin interaction with β cells of the pancreas is not yet clear. Fat cell-derived leptin can act as an inhibitory signal of osteocalcin activity in this feed-forward loop of insulin activity.
