The vascular endothelium provides natural anti-thrombotic effects by metabolizing adenine nucleotides (ATP and ADP) to generate the platelet inhibitor adenosine which acts on its G-protein coupled receptor to increase intraplatelet cAMP levels. Prostacyclin (PGI2) produced by endothelial cyclooxgenase (COX) also increases intraplatelet cAMP levels via the platelet IP receptor. The endothelium generates nitric oxide (NO), which at high levels inhibits platelet activation by generating cGMP. The platelet membrane proteins, PECAM, CECAM, ESAM, and JAM-A, attenuate responses possibly through homotypic interactions in closely adjoining platelets. Cx37 also negatively regulates platelet function, potentially by forming gap junctions between neighboring platelets and connexin-mediated exchange of cytoplasmic contents.
