More than 70% of the Runx3 RNA in stomach is estimated to be from epithelial cells. PCR product amplified from Runx3−/− mice is shorter than that from WT mice. Sequencing of PCR product confirmed the absence of exon 4 in RNA samples obtained from Runx3−/− mice (unpublished observation). Right, lanes 1–6 show PCR products amplified using primers specific for exon 3 (mRx3-exon3-F) and LacZ gene (LacZ-R; 5′-CTCTTCGCTATTACGCCAGC-3′). As expected, no bands were amplified from WT samples. For Runx3−/− samples, LacZ containing bands were amplified in lanes 4–6. The presence of these bands indicates the presence of LacZ-containing transcripts, which is presumably too few to elicit blue staining in GIT epithelium. X 30 and X 35 refer to the number of PCR cycles. A 100 bp ladder marker was used (M).
