The genome- and system-wide response of DNA methylation to early life adversity and its implication on mental health
Abstract
Early life adversity is associated with long-tem impacts on behaviour and physical and mental health. The mechanisms mediating the impact of early life environment on the phenotype are proposed to involve a change in the state of deoxyribonucleic acid (DNA) methylation and, as a consequence, in the stable programming of gene expression. Recent studies suggest that the changes in DNA methylation affect broad genomic regions, as well as peripheral tissues in addition to brain regions. Although the data are still scarce, it points to the possibility that DNA methylation is a mechanism of genome adaptation to signals from early life social environment. This modulation of the DNA methylation pattern is proposed to result in long-term impact on the phenotype that could become maladaptive under certain contexts later in life. This model has implications on our understanding of behavioural and mental health pathologies, as well as their diagnosis and therapeutics.
L’adversité en début de vie est associée à des effets à long terme sur le comportement et la santé mentale et physique. Il est proposé que les mécanismes facilitant l’effet de l’environnement de début de vie sur le phénotype opèrent un changement dans l’état de la méthylation de l’acide désoxyribonucléique (ADN) et, par conséquent, dans la programmation stable de l’expression génétique. Des études récentes suggèrent que les changements de la méthylation de l’ADN touchent de larges régions génomiques, ainsi que des tissus périphériques en plus des régions cérébrales. Bien que les données soient encore rares, elles indiquent la possibilité que la méthylation de l’ADN soit un mécanisme d’adaptation génomique aux signaux de l’environnement social du début de vie. Cette modulation du modèle de méthylation de l’ADN est proposée d’entraîner un effet à long terme sur le phénotype qui pourrait devenir mésadapté dans certaines circonstances plus tard dans la vie. Ce modèle a des implications sur notre compréhension des pathologies du comportement et de la santé mentale, ainsi que sur leur diagnostic et leur traitement.
