Abstract

Interference microscopy makes it possible to get information about the actual surface structure of the leaf. Transparent replicas were made using gelatin emulsions or plastic films which were examined with the Zehender chamber. From the interference picture a quantitative determination of the vault and pattern of the epidermal surface can be obtained. Interference microscopy with reflected light is a quick and reliable way to obtain quantitative records of the variety of surfaces. It can be used for recording the development and the variation of the surface layer under different physiological conditions. The method can also be applied for the detection of residues of pesticides.
