R-loop formation by Cascade and Cas9. Overall architectures of Cascade (Left) and Ca9 (Right) ribonucleoproteins and their respective R-loops. Although both complexes initiate R-loop formation by recognizing their respective PAM sequences flanking the protospacer, the Cascade reaction proceeds through a metastable intermediate, which is then locked to form a stable R-loop.
