SVG cells and their derivatives are free from JCV and BKV contamination. 5F4, SVG, 10B1, and HeLa cells were harvested in SDS lysis buffer 1, 2, and 3 days after being seeded. SVG p12 cells were harvested in SDS lysis buffer 2 days after being seeded. DNA was proteinase K treated, isolated by phenol-chloroform extraction, and tested for the presence of BKV DNA by quantitative PCR. No BKV DNA was detected in the intracellular or supernatant fraction of isolated DNA from SVG, 5F4, 10B1, or HeLa cells. In SVG p12 cells, BKV DNA loads were 4.7 × 106 copies/ng total DNA in the cellular fraction and 1.2 × 1011 copies/ml supernatant. Serial dilutions of the pM11 plasmid of the BKV Dunlop strain served as a standard curve and positive control.
