CNS2 protects Treg cell identity. Methylated CNS2 in Treg precursors prevents it from interfering with normal Treg differentiation. Demethylated CNS2 interacts with Foxp3 promoter in a T cell receptor (TCR) and NFAT dependent manner to promote and stabilize Foxp3 expression when mature Tregs are activated and exposed to diverse signals that shape Treg function.
