Abstract

Mechanisms establishing and maintaining promoter-enhancer interactions in hematopoietic stem cells (HSCs) to maintain stem cell identity are not fully understood. In this issue of Cell Stem Cell, Aranda-Orgilles et al. (2016) describe a role for a member of the Mediator complex in maintaining HSC-specific enhancers and hematopoietic homeostasis.

Aranda-Orgilles et al. propose that the Mediator complex member Med12 functions in enhancer regulation and survival of hematopoietic stem cells (HSCs). The loss of Med12 in HSCs (right panel) leads to loss of p300 binding, PolII, and H3K27ac at enhancer and super-enhancer regions that control key HSC genes. This leads to downregulation of gene expression and induction of HSC apoptosis, causing rapid bone marrow failure.
