IL-1α signaling in acetaminophen-induced liver injury. Acetaminophen overdose results in hepatocyte damage and DAMP release, which causes IL-1α release by Kupffer cells in a TLR4/MyD88-dependent manner. IL-1α then signals to neutrophils through IL-1R and its adaptor, MyD88, potentiating liver injury and the inflammatory cascade. DAMP, damage-associated molecular pattern.
