Abstract

In this issue of Cell, two articles show that tumor-specific changes in HLA-mediated antigen presentation affect tumor immunogenicity and may play a role in shaping cancer cell survival.

(A) Mutated cancer proteins are presented as neoantigens via the HLA-containing MHC class I complex to CD8+ T cells, triggering destruction of the transformed cell in a process called immunoediting. (B and C) Mechanisms of loss-of-antigen presentation, leading to cancer cell survival either by incompatible binding between the transformed protein and the individual’s HLA (B) or by loss of HLA heterozygosity (C).
