Doc2b sequesters Munc18 to facilitate SNARE complex formation and insulin secretion. Individual SNARE proteins are anchored in the two membranes. t-SNARE proteins, syntaxin and SNAP25 reside in the plasma membrane, and v-SNARE protein, VAMP, resides in the insulin granule membrane. Munc18 proteins bind the t-SNARE and v-SNARE proteins with high affinity, inhibiting SNARE complex formation. Doc2b competes with SNARE proteins and binds to Munc18 proteins, releasing the t-SNARE and v-SNARE proteins for complex formation, allowing granule fusion with the plasma membrane and insulin secretion. t-SNARE, Target membrane SNARE; v-SNARE, vesicle-associated SNARE; SNAP25, synaptosomal-associated protein 25; VAMP, vesicle-associated membrane protein; Doc2b, double C2-domain protein β.
