Upper panel: Integrated computational and experimental workflow to identify cancer associated lncRNAs using a cell cycle-based S-phase specific nascent RNA capture assay followed by transcriptome sequencing. Short Time-series Expression Miner (STEM) analysis identified S-phase specific temporally expressed lncRNAs. S-phase lncRNA expression and their methylation patterns in TCGA pan-cancer dataset. Lower panel: High-throughput clinical investigation on S-phase cancer associated lncRNAs (SCATs). Loss-of-function and gain-of-function experiments reveal FGF/FGFR dependent regulation cancer cell hallmarks. SCAT7 as a potential therapeutic target in cancer treatment as demonstrated using xenograft and PDX models. Model depicting the mode of action of SCAT7 in regulating pro-survival pathways.
