Overall summary of the findings by Li et al. (8). Shown are the generation of nascent hematopoietic stem cells (HSCs) and primitive MΦs in aorta gonad-mesonephros (AGM). The MΦs that express vascular cell adhesion molecule-1 (VCAM-1) migrates to the FL where they establish a niche in the vascular region to wait for the emerging HSCs with VCAM-1 receptor, VLA-4. The interaction between nascent HSCs and MΦs causes the HSCs to be retained in the fetal liver and this gives them a high probability to generate definitive hematopoiesis in the fetal liver. Since hematopoietic activity was noted in the bone marrow, there remains a question on the role of the VCAM-1+ MΦs in bone marrow hematopoiesis. ?, the unresolved path of MΦs to the FL.
