Autocrine and paracrine effects of galectin-1. As an intracellular scaffold protein, galectin-1 stabilizes Ras and Ras effector interactions, leading to more efficient Ras activation and greater tumor cell growth. In addition, galectin-1 increases CXCR4 expression, which further enhances chemokine (CXCL12)-mediated cell survival. As a paracrine factor, galectin-1 production by tumor cells reprograms non-neoplastic (stromal) cells in the tumor microenvironment, including T cells (immune regulation), endothelial cells (angiogenesis), and other cells types (e.g., macrophages) to secrete mitogens and chemokines that increase Ras and Ras effector (MEK) activation through receptor tyrosine kinase (RTK) and G protein-coupled receptor (chemokine) receptors (e.g., CXCR4), respectively.
