Stalled replication fork activates the FA pathway through ATR-mediated phosphorylation of FANCI. Phosphorylated FANCI, disrupts FANCD2 homodimer to form a heterodimer complex, which subsequently binds to ICL. FA core unit mono-ubiquitinates FANCD2, triggering conformational changes in the ID complex from an open channel to closed clamp.
