Contrast variation using small-angle neutron scattering (CV-SANS) allows investigators to veil the scattering contribution of component parts in a biological assembly. This is illustrated in this schematic: blue components represent species that disappear when normal buffers (0% D2O, left column, white background) are shifted to a 42% D2O mixture (right column, blue background). In the upper panels, nondeuterated protein machinery is generalized as blue mechanical gears, whereas the PAN-20S assembly in the lower panel is depicted as a blue meat grinder. By achieving this specific contrast, the only protein signal observed is that from the deuterated protein, shown in green. To see this figure in color, go online.
