Ca2+ efflux via TRPML1 channels in late endosomes and lysosomes creates a Ca2+ microdomain in the vicinity of neighboring RyR2 channels on the SR that triggers Ca2+ sparks via CICR. Consequently, the initial TRMPL1-mediated Ca2+ release event is amplified by RyR2s to generate a Ca2+ spark that activates BK channels on the plasma membrane.
