Transcription of MIR loci is positively regulated by MED complexes. Pri-miRNAs are processed by DCL1-centered microprocessors to produce miRNAs, which are in turn exported from the nucleus through AGO1-RISC complex. Cambiagno and colleagues found that, through the interaction with MED complexes and DCL1, HST recruits DCL1 to MIR loci and facilitates the capture and processing of pri-miRNAs. HST is imported from cytoplasm to nucleus by IMPA-2 and exported to cytoplasm by RAN1 without mature miRNAs. The bona fide cargos of HST remain to be identified.
