AAV research and production. Top part of the figure shows the three main areas in AAV research. i) AAV production – the goal is to produce AAV batches of high quality and purity while keeping the process time and cost-effective. ii) AAV cassette – where the end goal is to find cell-specific enhancer-promoter combination as well as efficient termination signals. iii) AAV capsid – AAV development is mainly focused on finding new cell-specific capsids or capsids with unique properties. However, increased packaging capacity and evading immune responses are also of great interest to the field of gene therapy. Bottom part of the figure shows a colour scale from green (Chloroform) to purple (Iodixanol) and for which experiments each method is more suitable. AAV: Adeno-associated virus.
