The pipeline utilized to understand the differences between mRNAs translated in soma vs. dendrites. Glock et al. (2) used hippocampal slices to physically dissociate neuronal somata (green shade) and neuropil (yellow), a region that is enriched for neuronal dendrites. After doing both RNA-seq and Ribo-seq analysis for both regions, the authors defined neuropil- vs. soma-enriched mRNAs through differential expression analysis. This allowed the authors to observe structural differences in mRNAs that are preferentially translated in each compartment by determining differences in the coding sequence (CDS) and noncoding regions (UTRs) of these mRNAs.
