Workflow of anti-CD19 CAR-T cell treatment for the SLE patient. T cells from the SLE patient were enriched from apheresis products and lentivirally transduced for CAR expression. The transduced CAR-T cells were expanded in vitro. The patient received autologous CAR-T cell infusion after lymphocyte depletion. The efficacy of the CAR-T cells in vivo was evaluated according to the B cell number and CAR-T cell percentage at different time points. The clinical disease severity evaluation for the SLE included the levels of autoantibodies, soluble complements, and proteinuria and the overall SLEDAI scores.
