Double function of the ACPA-CD147 axis in NLRP3 inflammasome signaling in RA macrophages. Anti-citrullinated peptide/protein antibodies (ACPAs) from rheumatoid arthritis (RA) patients bind to the CD147-integrinβ1 complex on macrophages, which initiates two signals through its signaling pathways. In one pathway, the ACPA-CD147 axis primes the inflammasome signal through the PI3K-Akt pathway and results in high NLRP3 transcription. In the other pathway, the axis activates the inflammasome signal through the ATP-K+ efflux pathway and produces activated caspase-1 and IL-1β. Blocking the key molecule CD147 may stop these two signals to suppress the inflammasome
