PLX5622 inhibits bone marrow monocyte production, reducing neuroinflammation and CNS infiltration following WNV infection. Treating mice with PLX5622 21 days prior to WNV-infection and until 7 days post-injury significantly reduces 1) monocyte proliferation in the bone marrow and 2) the number of microglia potentially producing pro-inflammatory cytokines and chemotactic molecules. This results in a substantial reduction in the recruitment of immune cells into the CNS, including nitric oxide-producing monocyte-derived cells (MCs) and neutrophils, and interferon gamma (IFN-γ)-producing T cells, remarkably reducing neuroinflammation and the disease phenotype, highlighting a protective role for PLX5622 in infection and monocyte-mediated disease.
