Reduced natural killer T (NKT) (-like) cell numbers are connected to vascular and parenchymal fibrosis. (A) Cluster of differentiation (CD) 3+CD56+ NKT-like cell numbers in freshly isolated pulmonary arteries from patients with pulmonary hypertension associated with pulmonary fibrosis (PF-PH) or chronic obstructive pulmonary disease (COPD-PH) compared with healthy donor controls (*P ⩽ 0.05 as determined by nonparametric ANOVA followed by Kruskal-Wallis multiple comparisons test). (B) Vascular collagen deposition was determined by quantification of Masson trichrome stainings. Vessel was defined by outer media border (**P ⩽ 0.005 as determined by nonparametric ANOVA followed by Kruskal-Wallis multiple comparisons test). (C) Quantification of various NKT (-like) cell subsets in patients with ILD or COPD with unknown pulmonary hemodynamic status. Peripheral blood mononuclear cells (PBMCs) were isolated from freshly drawn whole blood by density gradient centrifugation. PBMCs were stained for cell surface markers, and the CD14-negative population of PBMCs was quantified (n = 5–6; **P ⩽ 0.005, *P ⩽ 0.05 as determined by nonparametric Mann-Whitney test).
