Ferroptosis of PMN-MDSCs causes immune suppression in cancer. Spontaneous ferroptosis of PMN-MDSCs in the tumor microenvironment triggers the secretion of PGE2 and the release of oxidized phospholipids, rendering them more immunosuppressive through influencing the activity of CD8+ T cells and tumor-associated macrophages. AA arachidonic acid, ACSL4 acyl-coenzyme A synthetase long-chain family member 4, Alox12/15 arachidonate 12/15-lipoxygenase, GPX4 glutathione peroxidase 4, PE phosphatidylethanolamine, MPO myeloperoxidase, PGE2 prostaglandin E2, PMN-MDSCs pathologically activated neutrophils termed myeloid-derived suppressor cells, TAM tumor-associated macrophage.
