A. After ligand-mediated activation of the βAR, the receptor is desensitized through phosphorylation by GRK and β-arrestin binding. The receptor is internalized in endosomes and then re-sensitized by dephosphorylation by PP2A. The receptor is subsequently returned to the plasma membrane where it can be activated again. B. Exposure to hypoxia leads to increased phosphorylation of internalized βAR and activation of PI3Kgamma. Active PI3Kγ phosphorylates the endogenous inhibitor of PP2A (I2PP2A) which then interacts with PP2A, inhibiting its activity. De-phosphorylation of βAR in endosomes is abrogated leading to inhibition of re-sensitization. Figure was created with Biorender.com.
