Nitration of Caveolin-3 (Cav-3) at Tyrosine 73 (Tyr73–3NT) induces insulin resistance and increases susceptibility to myocardial ischemia in prediabetic mouse hearts. ONOO is produced near Cav-3 in the presence of O2 and NO. Tyr73–3NT in the caveolin scaffolding domain (Cav scaff domain) prevents signalosome formation and disrupts transmembrane signaling through insulin receptors (InsRs), adiponectin receptors (AdipoRs) and other receptors. ONOO may be scavenged in the presence of superoxide dismutase (SOD), peroxiredoxin (Prx) or ebselen, a glutathione peroxidase mimetic.
