Thrombocytopenic post-natal day 14 (P14) mice lacking the Thrombopoietin (TPO) receptor were transfused with platelets obtained from either WT adult mice (top) or WT P7 newborn pups (bottom), and the effects of these transfusions on the recipient’s monocytes were evaluated. Ly6Chi (pro-inflammatory) monocytes increased significantly in response to adult as well as neonatal platelet transfusions. However, pups transfused with adult platelets exhibited significantly increased platelet-monocyte aggregates (PMAs) and trafficking monocytes (typified by increased CCR2 and CCR5 surface expression), compared to pups transfused with neonatal platelets. Addition of an anti-PSGL-1 antibody partially reversed these findings, suggesting that the transfusion of adult, but not neonatal, platelets to newborn mice induces a trafficking monocyte phenotype, at least partially through P-selectin mediated cell-cell interactions.
