Cytoplasmic DNA or Mn2+ stimulates cGAS aggregation and its enzymatic activity to catalyze ATP and GTP consumption for the synthesis of 2′3′-cGAMP. 2'3'-cGAMP binds to STING in the endoplasmic reticulum membrane and stimulates its translocation to the Golgi. In the trans-Golgi network (TGN), ARMH3 interacts with STING and recruits PI4KB to synthesize PI4P near STING. Furthermore, PI4P-binding proteins, such as AP-1 and GGA2, direct STING translocation to clathrin-coated vesicles (CCVs), which eventually traffic STING to endosomes, inducing STING activation. Other PI4P-binding proteins, such as lipid transporter proteins, facilitate lipid transportation and establish a lipid environment conducive to STING activation
