Figure.. PDCD5 (programmed cell death 5) mediates a negative feedback loop by HDAC3 (histone deacetylase 3) degradation.
(1) TGFβ (transforming growth factor-β)/SMAD (SMAD family member) signaling drives expression of PDCD5. (2) PDCD5 associates with HDAC3 in the nucleus to (3) promote export from the nucleus for ubiquitination and degradation. With loss of nuclear HDAC3, (4) disinhibited SMAD7 negatively feeds back to reduce/repress myofibroblast gene expression. (5) Previous reports of PDCD5 interacting with p53 may reinforce/promote myofibroblast senescence.
