Panel A shows the structure of complete primase-NTPase (OPG117) based on PDB entry 8APM (residues 321-784). The primase domain of OPG117 (1-321) was modeled using AlphaFold through ColabFold. The obtained protein model’s overlap region (321-400) was superposed onto the existing helicase structure region (321-400). Panel B shows a proposed RC consisting of four viral proteins. The structure of DNA polymerase is from the Protein Databank entry 8HG1 {Peng, 2023 #93}. The palm subdomain of the DNA polymerase subunit in 8HG1 was superposed onto the previously modeled structure of MPXV {Kannan, 2022 #94}. The template (cyan) and primer (orange) were modeled using the DNA-bound structure HSV pol (PDB entry 7LUF) and S. cerevisiae replicative polymerase δ. This figure shows that modeled template-primer superposes extremely well onto the one reported in the cryoEM structures (template – magenta, primer – gray) [4-6]. The 3’–5’ Exonuclease domain of DNA polymerase is colored in limon color, the fingers in blue, the N-terminal in light grey, the palm in red, and insert 3 in light pink. The predicted structure of OPG148 was superposed onto the reported structure of VACV OPG148 (1-50) bound to OPG71 (PDB entry 6XZP) to obtain a relative position of OPG148 and OPG71. OPG148 is shown in forest green color. Similarly, the OPG116 was modeled through homology modelling using VACV OPG116 bound to a small segment of OPG148 as a template (PDB entry 4OD8).
