Abstract

GABA signaling by melanoma cells was found by Tagore and colleagues to trigger keratinocyte-driven growth of melanomas. This study reveals new roles for nonneuronal signaling by a neurotransmitter in regulating tumor initiation and outgrowth. See related article by Tagore et al., p. 2270 (4).

Reciprocal signaling between melanoma cells and keratinocytes regulates melanoma growth. Findings from Tagore and colleagues support that GABA is synthesized in melanoma cells and secreted via exocytosis to keratinocytes. In keratinocytes, GABA activates GABA signaling and hyperpolarizes cells via chloride ion (CI) efflux, and these activities result in the activation of MYCN, which in turn promotes transcription of leukemia inhibitory factor (LIF). LIF is secreted by keratinocytes to regulate the proliferation and potentially other properties of melanoma cells.
