FTO (Fat mass and obesity associated) regulates inflammasome activation during acute lung injury. FTO, an RNA demethylase, promotes the expression of TXNIP (Thioredoxin interacting protein), a protein that binds NLRP3 (NOD-, LRR-, and pyrin domain-containing protein 3) and promotes inflammasome activation. (A) Under conditions of high FTO expression, inflammatory stimuli (such as LPS) lead to high TXNIP induction, inflammasome activation, and inflammatory cytokine production. (B) Inhibition of FTO expression prevents high TXNIP induction after LPS, leading to reduced inflammasome activation and cytokine production. m6A = N6-methyladenosine.
