Schematic representation of MMPs-mediated induction of arterial medial calcification. Inducers of vascular calcification (such as elevated phosphate and calcium levels) affect the expression and activity of several proteases. In particular, cathepsins and MMPs are activated and involved in extensive elastolysis and ECM remodelling. Elastin degradation and ECM disorganization then favour ectopic calcification in the medial layer of the arterial wall, which is a risk factor for CV events and mortality. Moreover, MMPs facilitate the initiation and progression of ectopic calcification by directly promoting osteochondrogenic differentiation of VSMCs. Figure created with BioRender.com.
