Single-cell and imaging mass spectrometry reveals alterations in the immune cell signature in alveolar tissue from patients with emphysema. (A) Freshly collected lung tissue from emphysematous or control lung tissue was digested to single-cell suspensions and incubated with myeloid and lymphoid panels of metal-labeled antibodies for analysis by single-cell mass cytometry. Frozen formalin-fixed, paraffin-embedded sections from emphysematous or control lung tissue were incubated with myeloid and lymphoid panels of metal-labeled antibodies for analysis by imaging mass cytometry. (B) Single-cell mass cytometry analysis revealed alterations in the immune cell signature in alveolar tissue from patients with emphysema versus control tissue, including increased percentages of central memory CD4 and CD8 T cells and CD1c+ dendritic cells. Spatial imaging demonstrated colocalization of CD8 T cells and CD1c+ dendritic cells in emphysematous lung tissue. Figure created with BioRender. FFPE = formalin-fixed, paraffin embedded.
