A metabolic target for Alzheimer’s
Amyloid-β peptide and tau protein activate the kynurenine (KYN) pathway by increasing indoleamine-2,3-dioxygenase 1 (IDO1), the enzyme that metabolizes tryptophan (TRP) to KYN, which impairs astrocyte metabolism, lactate flux, and neuronal function. Blocking IDO1 with PF068 or through genetic deletion restores metabolic flux, neuronal function, and cognition, demonstrating that glial metabolism is a potential therapeutic target in Alzheimer’s disease.
