Molecular Analysis of Positive Intradermal Tests and Lymphocyte Transformation Test. (A) Total RNA was extracted from skin biopsies, and gene expression of CD8A, CD4, GLNY, GZMB, CD69, IFNG, CXCL10, HLA‐DRA, GPR183, PLEK, IL‐15, and IGFL3 was analyzed by qRT‐PCR. Expression values were normalized to GAPDH. Results are expressed as fold changes relative to control skin. Und, Undetected (B) Patient peripheral blood mononuclear cells were isolated by Ficoll density gradient centrifugation and exposed to varying concentrations of the tested drugs or control drug (amoxicillin). Negative (drug‐free, white bar) and positive (non‐specific activators, gray bars) controls ensured experimental reliability. T lymphocyte proliferation was measured by [3H]thymidine incorporation; results are expressed as the proliferation index (ratio of 3H in drug‐stimulated to unstimulated cells). Three healthy donor controls were tested negative for iomeprol and iodixanol (five increasing drug concentrations respectively, not shown).
