Abstract

In this study, using chitosan (CS) and β-cyclodextrin (β-CD) as raw materials, a cyclodextrin grafted chitosan (CD-g-CS) derivative was successfully synthesized. A novel CD-g-CD-SA-based thermosensitive hydrogel was developed by physical crosslinking in the existence of sodium glycerophosphate (GP) using CD-g-CS and sodium alginate (SA) as materials. The gel time of hydrogel was short, which was <5.00 min. The results of SEM showed that the hydrogel had porous reticular structures. In vitro swelling test and lysozyme degradation experiment demonstrated the good swelling and biodegradability of hydrogel. Using berberine hydrochloride (BBH) as the model drug, the hydrogel showed a slow and controlled release over 36 h and the cumulative release of 36 h was 59.87 %, which demonstrated the hydrogel had good drug release effects. The antioxidant experiments showed that hydrogel had a good antioxidant capacity. The results of the hemocompatibility test and the cytotoxicity test confirmed that the hydrogel had good biocompatibility and biosafety. In vitro antibacterial tests proved that the hydrogel had significant antibacterial effects on both Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). In summary, the CD-g-CD-SA/GP hydrogel has great potential for development in the field of biomaterials such as drug sustained release and bacteriostatic materials.
