Abstract

This chapter presents a protocol for short tandem repeat (STR) analysis, a robust, economical, and highly accurate method commonly used in forensics and agriculture for genetic profiling and identity determination. The protocol involves DNA isolation from FTA cards or leaf pieces, followed by PCR amplification of STR regions and fragment analysis. The number of repeats in these short tandem patterns is unique to individuals, making it useful for genotyping analysis. By analyzing STR profiles from reference samples, a reference STR table is generated, serving as a comparison tool for identifying repeat differences between the genotypes of test individuals. This comprehensive analysis of genetic variations provides valuable insights for differentiating between individuals based on their unique genetic profiles.
