Figure 1:. GPR146 overexpression in VSMCs leads to Piezo1 transcription to induce hypertension.
GPR146 is upregulated under HHP, which causes Gs-cAMP-PKA-CREB phosphorylation, translocation to the nucleus, and Piezo1 upregulation. GPR146 overexpression also upregulates TRPV4 and TRAAK channels, culminating in synthetic VSMCs, vascular remodeling, and hypertension. Blocking GPR146 attenuates blood pressure, maintains VSMCs in the contractile phenotype, and lowers circulating cholesterol. This represents a promising novel therapeutic approach for hypertension and hyperlipidemia (created with Biorender.com).
