Abstract
Contra-thermodynamic stereochemical editing represents an innovative strategy that overcomes thermodynamic barriers, enabling the conversion of a set of chiral centers from their thermodynamically stable configurations to high-energy stereochemical configurations. Energy compensation and chirality enrichment could be the key challenges in the design of such reactions. This Perspective highlights significant advancements in this realm, focusing on the mechanisms that drive these processes through the integration of various energy and chiral sources. The discussion covers several reaction types, including autoamplification, deracemization, and epimerization.
